Description
Chloramphenicol is a sterile otic suspension containing Chloramphenicol 0.5% in Hydroxypropyl Methylcellulose to improve spreadability, absorption, and penetration.
Each mL contains: Active: Chloramphenicol 5 mg.
Excipients/Inactive Ingredients: Vehicle: Hydroxypropyl Methylcellulose 5.0 mg (4,000 cps).
Preservative: Thimerosal 0.1 mg.
Inactive: Boric Acid 20 mg, Sodium Borate (to adjust pH); Purified Water.
Action
Pharmacology: Chloramphenicol is a broad-spectrum antibiotic originally isolated from Streptomyces venezuelae. It is primarily bacteriostatic and acts by inhibiting protein synthesis by interfering with the transfer of activated amino acids from soluble RNA to ribosomes. Development of resistance to Chloramphenicol is minimal for staphylococci and many other species of bacteria.
Indications/Uses
For the treatment of chronic suppurative otitis media, otitis externa, chronic otorrhea, infections of fenestration and mastoid operation cavities and other ear infections caused by Gram-positive and Gram-negative organisms including Staphylococcus aureus, E. coli, H. influenzae, Aerobacter aerogenes, Klebsiella pneumoniae, and Proteus species.
Dosage/Direction for Use
Instill two drops into the external auditory canal. If necessary, a wick can be soaked with 4-6 drops and packaged loosely into the infected cavities but should be renewed every 12-24 hours.
Contraindications
Contraindicated for patients who have previously shown sensitivity to the drug.
Warnings
Prolonged or frequent intermittent use of topical Chloramphenicol should be avoided because of the possibility of hypersensitivity reactions, including bone marrow hypoplasia.
Special Precautions
In all except very superficial infections, topical Chloramphenicol should be supplemented by appropriate systemic medication. Overgrowth of non-susceptible organism may occasionally result. Appropriate measures should be taken.
Special Precaution: Otic preparations containing antibiotics should be used with caution in cases where the tympanic membrane is perforated due to ototoxicity risk.
Storage
Keep tightly closed after use. Store at temperatures not exceeding 30°C.
